The experiment was about optimizing PKC fermentation process by Trichoderma reesei, was started with growth characteristic observation of colonies number of Trichoderma reesei, with the equation as follows Y= 0.1745x2 + 2.2611x – 0.4699. The optimum amount of 2,13 x 106 CFU/cc Trichoderma reesei was reached at 60 hours. The highest percentage of colony diameter was reached at 30 hours that was 102.89%. Completely Randomized Design with factorial pattern 3x3 was used in this second phase. The first factor was the thickness of PKC medium (1, 2, and 3 cm) and the second factor was Trichoderma reesei doses (104, 105, 106 CFU/cc). There was a significantly different ( p<0.05) of Trichoderma reesei dose and thickness of medium on pH and temperature medium, ADF, NDF, and hemi cellulose content, but not significant on dry matter. There was positive interaction between thickness of medium with T. reesei dose on NDF, hemi cellulose content of PKCF.
